About the Job

The Hormel Institute at the University of Minnesota is seeking a Research Specialist to join the CryoEM facility. This role supports the local research community at The Hormel Institute, as well as external academic and industry partners. The position works under general supervision and exercises discretion to make daily operational decisions based on best practices.

Focus of Job Responsibilities

Research Design and Operation of Associated Instruments & Protocols - 50%

* Manage and optimize use of the Arctis
* Develop workflows that links sample preparation (Arctis) to data collection (Krios).
* Develop and optimize sample and data collection parameters.
* Develop protocols for SEM and IFLM sample screening, milling of vitrified samples, waffle milling and cryo sectioned samples.
* Optimize protocols for slice and view function.
* Operate Titan Krios as needed and provide support for tomography data collection.
* Prepare grids and reagents for ET experiments including carbon coated grids for negative stain, graphene/continuous carbon grids for cryoEM, negative stains and fiducials.
* Perform conventional vitrification using the Vitrobot, CP2, Leica GP2 and slammer.
* Culture live cells on grids for cryoET experiments, apply adhesive coatings to grids for cell culture, and prepare grids for cell culture via micropatterning. Additionally, develop protocols for frequently used cell lines and making a library of preferred conditions.
* Perform live cell vitrification using both conventional vitrification instruments, and high pressure freezing for standard FIB-milling and the Waffle technique.
* Perform vitrification of tissue samples using a high-pressure freezer prepared through various means such as microtome.
* Troubleshoot experimental approaches for a variety of different samples and experiments to meet users needs.
* Instrumental operation including image acquisition and microscope maintenance.
* Implement data collection strategies that will benefit our user community.

Problem Resolution - 25%

* Exercise considerable resourcefulness to find solutions to research issues within budgetary constraints.
* Applies subject matter knowledge, including research technologies, to ensure operational effectiveness.
* Lead workshop development and functions.
* Assign duties to technical support staff.
* Demonstrate decision-making that is guided by policies and procedures created by the cryo-EM team.
* Serve as a liaison with internal and external contacts regarding research.
* Modify policies and procedures to improve efficiency and quality.
* Proactively search for a better way to effectively achieve end results.
* Achieve a scope of measurable impact.

3D Data Processing, Reconstruction, & User Support - 25%

* Collect and process data for 3D map and tomogram reconstruction.
* Produce subtomogram averages and segmentations as needed.
* Provide experimental and analytical support to users.
* Engage in data analysis including image analysis, data processing and reconstruction.
* Perform single particle analysis reconstruction using CryoSPARC and Relion.
* Perform tomogram reconstruction, segmentation and subtomogram averaging using WarpTools, Imod, Aretomo and Chimera/ChimeraX.
* Engage with users to train and provide experimental support in sample preparation such as negative stain, culturing cells on grids and vitrification.
* Operate instruments such as Vitrobot, High Pressure Freezer and Arctis, Krios.
* Analyze data through tomogram reconstruction, segmentation, subtomogram averaging and single particle analysis.
* Provide support in experimental set-up or considerations for single particle and tomography experiments.
* Serve as the lead on assigned projects.

Some on-site work may be necessary for certain positions, even those designated as fully remote. This position has been designated as eligible for Flexible Work Profile One.
Flexible Work Profile One is expected to work primarily on-site. Your office will be located at The Hormel Institute in Austin, MN. The Hormel Institute management retains the right to modify flexible work arrangement agreements on a temporary or permanent basis for any reason at any time.

Qualifications

Required Qualifications

* Ph.D., DDS, DVM, JD, MD or equivalent
* Proven experience with fundamental biochemistry and molecular biology techniques.
* Experience in structural biology, with emphasis on cryo-electron tomography.

Preferred Qualifications

* Ph.D. in chemistry or biochemistry preferred.

Pay and Benefits

Pay Range: $81,037- $92,232.28; depending on education/qualifications/experience

Time Appointment Category: 75%-100% Appointment (If job description above does not specify appointment or expected weekly hours, assume a 100% appointment.)
