Gene sequence analysis, vector design, vector construction and cloning, vector QC, potentially also dicot and/or monocot transformation, editing sequence analysis, greenhouse plant care.
Tasks include:
- Defining the promoter, CDS, intron and terminator sequences needed to optimize gene expression of genome editing components in vectors built for diverse monocot and dicot crops.
- Designing the gene synthesis, vector construction / cloning strategy, and executing on that strategy through directing the work of post docs or PhD scientists in the lab
- Identifying and ordering the right equipment to set up the lab (with consultation from the PI)
- Defining the transformation and tissue culture conditions appropriate for transforming and regenerating specific specialty crops and row crops, for both monocots and dicots, working with PI and the Senior Research Scholar Dr. Kedong Da.
- Identifying the promoter, CDS, intron and terminator sequences needed to optimize gene expression of recombination technology and morphogenetic factors to enhance regenerability, with an emphasis on synthesizing knowledge from anecdotal data / experience, with literature, and personal contacts within other organizations
- Confirming vector designs are correct and stabilizing our vector construction protocols for future ease of use by new students
- Creating protocols from those processes for simplified training procedures
- Collaborating across research projects in diverse departments who are working with the GEC on the vector design strategies
- Designing genome editing trait constructs including optimizing gRNA structure, specificity, cross-germplasm homology, and minimizing off-target effects
